1) This is double stranded. Won't it either cause apoptosis or RNA silencing?
The RNA molecule
is blocked with an oligo. The oligo can be DNA or modified RNA. A DNA-RNA complex does not induce apoptosis or RNA silencing.

This is RNA so will it be degraded before the oligo is removed by wt RNA?
It depends. The half-life
of RNA is on the order of minutes to hours, but the competition reaction is on the order of seconds to minutes, so if wtRNA
is present, the oligo will be competed off before the molecule is degraded depending on the concentrations used. Also, the
Nucline construct can be stabilized to increase half-life using any number of methods including adding 3' UTR stabilizing
sequences and using chemically modified oligos that block 5' ribonuclease activity.
3) How does the oligo come
off in target cells?
Through competitive hybridization and strand displacement. In a target cell, the oligo has a
choice-- stay bound to Nucline or bind to the wtRNA. When the target wtRNA is present, the oligo is competed off Nucline and
the reaction proceeds to equilbrium. In non-target cells, the oligo stays in place until Nucline is degraded.

What does a license cost?
Right now, nothing to use the molecule for research. A simple "use" license is available for download. Please
see below and send it back. A person or organization(for-profit or not) can make and use Nucline under this license but cannot
sell, bulk manufacture, import, or export the molecule. We do not ask for or restrict how the data is used under these licenses.

5) How long is
the license good for?
Two years from the day we receive it. Beyond that, a new license is required.

What to I get for signing up?
Of course the legal protection that licenses provide, but also unlimited technical support, and
in the event the "use" license is withdrawn, one month's notice prior to withdrawal and the ability to continue to use Nucline
for the full term of the contract until it naturally expires.
